Petersen was withdrawn from last week`s Test in Port Elizabeth after picking up a virus, although he has since recovered and should open the batting alongside skipper Graeme Smith, with Dean Elgar taking De Kock`s place in the middle order.
According to Sport24, De Kock scored seven and 34 in Port Elizabeth, where he debuted, batting in the unaccustomed position of No 6 and did not keep wicket while Elgar, who opened in place of Petersen, scored 83 and 16 in his two knocks at St George`s Park.
Meanwhile, spinner Robin Peterson is likely to miss out again with the hosts expected to go with a four-man pace attack, added by the spin-options of JP Duminy and Elgar.
With left-arm seamer Wayne Parnell out with a groin strain, either Rory Kleinveldt, Kyle Abbott or Ryan McLaren will get a chance, the report added.
